Modified Methods of Hookworm Culture, Especially for the Continuous and Massive Culture.

Abstract
Often enormous numbers of hookworm larvae are required for some research projects. The filter paper-test tube method has been considered more convenient than the classical charcoal method for the hookworm culture when small amounts are used. However, in the case of ;large ¢¥size cultures, there is considerable disadvantage with both of these methods.
Two new methods of bookworm culture were tried. In one method the filter paper is attached inside of a U-shaped plastic plate and put on the media-water in a fl-shaped manner (UI). The other method was that of using a particular pottery in which the surface was specially smooth and a good absorbent of water (PT). These two methods were compared with the filter paper-test tube method the cast plate method, the architectural tile method, and the charcoal method. Among these variousculture methods, the UI and the PT method were the most efficacious and convenient. Also the PT method gave,.maximum numbers of larvae and at an earlier time than with the filter paper-test tube method and also a considerable number of rhabditoid larvae were obtained by soaking the plate thoroughly in warm water after one or two days of culture (Fig. 6, Fig. 7).
Therefore, the two methods (PT & UI) are considered ¢¥to be convenient and efficacious for the following reasons; 1) A greater number of specimens can be set in a limited place. 2) The culture and collecion of larvae is simple. 3) Easier change of medium-water in these methods prevents the hatched larvae from expiring in the media-water. 4) The culture material is durable, and wastage is negligible. 5) Especially in the PT method, the collection of rhabditoid larvae is done conveniently and efficaciously.
It is suggested that the U-shaped plate (UI) should be made in a rectangular form and its longer side be put on the water for efficiency, and the pottery plate (PT) should be made L-shaped for convenience.
